Many startups emerged since the advent of the fourth generation of AI and its extensive and rigorous developments. Even a small startup could reach a candidate in a couple of years and AI-discovered molecules have been brought into clinical trials. Biotech companies specialized in AI (AI-native biotech) could change the scenario of drug discovery. But there is a concern on AI-discovered drugs that comes from the unprecedented nature.<\/p>\n<p>This perspective article reviewed the AI-discovered drugs in clinical trials.<sup>1)<\/sup> There has been no approved AI-designed drug until 2023. It is hard to say that they go through the whole process to reach the market at that moment, but their analysis on the clinical pipelines of AI-native Biotech companies indicated positive outcome.<\/p>\n<p>AI-discovered drugs, which mean AI played a major role in drug discovery, are classified to several categories. So far candidates for AI-discovered targets and AI-designed small molecules are the major molecules in clinical trials. Antibodies , vaccines and repurposed molecules are also in clinical trials and the numbers are gradually increasing.<\/p>\n<p>AI-discovered candidates have better success rate in phase I than conventionally developed ones. Among the 64 candidates that entered Phase I clinical trial, 24 have been finished and 21 compounds were successful to cross the safety criteria. AI-repurposed molecules, All the AI-designed antibodies and vaccines passed through phase I. The success rate of AI-designed small molecules, was 67% (2 out of 4), but that of AI-discovered targets was 90% (9 out of 10). Even though the data in phase II is scarce at that moment, 4 out of 10 candidates are successful.<\/p>\n<p>The high success rates in clinical trials so far indicate that AI-first discovery is capable of identifying druggable molecules and targets. The analysis is just a prompt report on AI-driven drug discovery but it suggests that it is worth taking continuous efforts on application of AI technologies.<\/p>\n<p>The first AI-designed drug candidate was from Exscientia in 2020 and the followers have thrown their pipeline molecules into clinical trials during the past 5 years. It is amazing that 67 candidates have entered until 2023. Although it is better to take a close look at the coming results of further progress in clinical, there is nothing particular to doubt the AI-discovered molecules.<\/p>\n<p>This early-stage analysis on AI-discovered drugs provides us insights on the way for the application of AI. AI-first drug discovery is possible but the success rate of AI-designed molecules was lower than other categories. It is, at least at this moment, better not to fully depend on AI.
