Mechanism-Based Evaluation of Protein–Protein Interaction Inhibitio
To advance drug discovery programs targeting protein–protein interactions (PPIs), PRISM BioLab has established in-house in vitro pharmacological assays. Using purified proteins or cellular systems, these assays directly detect and quantify PPI inhibition by compounds.
A broad range of complementary assay technologies—including HTRF, FRET, fluorescence polarization (FP), NanoBiT, Alpha assays, and CETSA—are combined to design optima evaluation systems depending on the target PPI and research objectives.
This integrated approach enables consistent assessment from direct interaction inhibition using purified proteins to validation of intracellular PPI inhibition and confirmation of target engagement in cells. By not relying on a single assay but instead using orthogonal methods, PRISM BioLab reduces false positives and negatives, thereby enhancing confidence in mechanism-of-action assessment.
These data are used for hit selection, analog optimization, and progression to subsequent functional assays, supporting sustained generation of high-quality candidate compounds with well-defined mechanism of action.
